Amlodipine besylate
Indication
1st-line treatment for HTN as monotherapy or combination w/ other antihypertensive agents; myocardial ischemia, whether due to fixed obstruction (stable angina) &/or vasospasm/vasoconstriction (Prinzmetal's or variant angina) of coronary vasculature as monotherapy or combination w/ other antianginal drugs.
Instruction
May be taken with or without food. HTN & angina Initially 5 mg once daily. Max dose: 10 mg daily.
Drug interaction
Increased systemic plasma concentration with immunosuppressants (e.g. ciclosporin, tacrolimus). Increased serum concentration of simvastatin. Increased exposure with CYP3A4 enzyme inhibitors (e.g. protease inhibitors, azole antifungals, erythromycin, diltiazem). Decreased plasma concentration with CYP3A4 inducers (e.g. rifampicin).
